Oral delivery of macromolecules, including peptides and proteins, remains challenging due to the harsh gastrointestinal environment, rapid enzymatic breakdown and limited permeability across biological membranes. Emerging technologies such as advanced lipid-based formulations, delayed-release softgels and buccal or sublingual systems are helping to address these barriers. By protecting compounds, enabling targeted intestinal release and utilizing lymphatic transport pathways, these approaches aim to improve systemic exposure. This presentation will review the scientific principles and supporting data behind these innovations, along with an overview of the current landscape and recent progress in oral macromolecule delivery.
