Lonza has developed a platform that allows dual-payload antibody-drug conjugates (dpADCs) to be made accurately and reliably. With existing technologies, it is difficult to ensure a consistent drug-antibody ratio for both of the payloads. The Lonza approach is an extension of its proven GlycoConnect® technology which, in combination with its HydraSpace® spacer technologies, allows dpADCs with high homogeneity to be created reliably and repeatably. It is applicable across a broad spread of different payloads, and allows both ratios and stoichiometries to be precisely tailored. Importantly, it produces CMC-ready processes that make stable, homogeneous products, and there has already been success in preclinical studies with a dpADC made in this way. dpADCs offer the prospect of more targeted therapy while also, potentially, avoiding the resistance that is all too common in cancer treatment. Being able to design and make them accurately and reliably represents a huge step forward for this new class of molecules, and Lonza’s technology moves commercialized dpADCs closer to being a reality.
