16mg/5mg/12,5mg ;16mg/10mg/12,5mg capsules, FDC of the ARB candesartan cilexetil, the dihydropyrine CCB amlodipine and the diuretic HCTZ is clinically meaningful because the monocomponents contribute to the antihypertensive effect by complementary mechanisms of action:fi •the selective and competitive binding of candesartan to the AT1 receptor prevents binding of angiotensin II and its vasoconstrictor response, •amlodipine by inhibiting L-type calcium channels in vascular smooth muscle cells exerts a direct vasodilatory effect, •HCTZ by inhibiting the renal sodium reabsorption induces diuresis and on the long-term reduces total peripheral resistance.
